    采用34个微卫星分子标记,对长江(湘江、监利、枞阳)、珠江、黑龙江(抚远)鲢共5个群体的观测杂合度(Ho)、期望杂合度(He )、多态信息含量(PIC)和有效等位基因数(Ne)等进行了遗传检测,根据基因频率计算遗传相似系数和Nei氏标准遗传距离,以卡方检验估计Hardy-Weinberg平衡,以近交系数(FST)和基因流(Nm)分析群体的遗传分化。同时,基于Nei氏标准遗传距离获得NJ聚类图。共检测出154个等位基因,其中有80个共有基因。位点等位基因1~9个不等,平均等位基因4.666 7,平均有效等位基因2.948 9。5群体平均观测杂合度为0.329 5~0.461 9,平均期望杂合度为0.500 2~0.552 9,平均多态信息含量为0.443 5~0.493 0。表明5群体为中度多态,遗传多样性较低,且长江不同地理群体的遗传多样性也有差异,但差异不显著。通过群体间遗传相似性和遗传距离比较显示:长江3个群体间遗传相似系数最大,遗传距离最小;黑龙江与长江鲢群体间相似系数较大(平均0.871 8),遗传距离较小(平均0.137 4)表明二者遗传分化亦最小;珠江鲢与长江鲢间的相似系数最小(平均0.831 0),遗传距离最大(平均0.185 2),其遗传分化差异较大,结果未显现出群体间遗传差异的大小与其地理距离远近呈正相关。

    Abstract:

    In this paper, genetic diversity of five wild silver carp(Hypophthalmichthys molitrix)populations, collected from the Yangtze River, Pearl River and Heilongjiang River was analyzed using 34 microsatellite markers. The observed(Ho) and expected heterozygosity (He),polymorphism information content(PIC) and number of effective alleles(Ae) were all determined.The genetic similarity coefficient and Nei'S standard genetic distance were computed based on the allele frequencies.The HardyWeinberg equilibrium was checked by X2 test.Genetic differentiation and hierarchical partition of genetic diversity were evaluated by FSTand Nm.A dendrogram was constructed based on NJ method using MEGA3 software package .For each locus, 1-9 alleles were amplified with an average of 4.666 7,adding up to 154 alleles in all five populations, of which the mean valid alleles were 2.948 9.We found the genetic variability level was moderate in the five populations, as shown by Ho(0.329 5-0.461 9),He (0.500 2-0.552 9) and PIC(0.443 5-0.493 0),respectively. Based on the genetic similarity coefficients and genetic distance, the genetic diversity between the two groups of Yangtze and Pearl Rivers(Ds=0.1852) was higher than that of the two populations of Heilongjiang and Pearl Rivers(Ds=0.1558). There were no obvious relations between genetic distance and geographical distribution of the five populations.
