    • Molecular cloning and sequence analysis of insulin-like growth factor I cDNA from Ctenopharyngodon idellus

      Abstract:Total RNA was isolated from grass carp ( Ctenopharyngodon idellus ) liver tissue. The cDNA encoding insulin-like growth factor I ( IGF-I) peptide was amplified by reverse transcript ion polymerase chain reaction ( RTPCR) strategy using isolated total RNA as template. The amplif ied cDNA fragment was inserted to vector pUC18. The cloned cDNA was sequenced and the amino acid sequence of grass carp IGF-I was predicted. The nucleotide sequencing showed the cDNA encoded 117 amino acids including B, C, A, D and E f ive domains. The nucleotide sequence and deduced amino acid sequence shared 93. 8% and 97. 1% homology with that of common carp mature IGF-I. Analysis of E domain indicated that cloned grass carp IGF-I belonged to IGF-IEa-2 subtype.

    • Partial isolation and bioassay of gonad2stimulating hormone from the brain of Macrobrachium rosenbergii

      Abstract:The extracts of brain of Macrobrachium rosenbergii were applied on a Sephadex G -50 column (1. 6cm ×50cm) . Two large peaks of protein , peak Ⅰand peak Ⅱ, were obtained. The ⅡC(63 -74 fractions) of peak Ⅱwere able to increase oocyte diameter of M . rosenbergii in vitro very significantly. The 63 -65 fractions from ⅡC of peak Ⅱ significantly accelerated the total protein synthesis of the prawn ovary in vitro. By injecting the 63 -65 fractions into muscle of M . nipponense the ovary maturation was promoted significantly after 15 days. The molecular weight of the protein in the fraction 63 was below 7kD. The results show that the gonad2 stimulating hormone is present in the brain of M . rosenbergii.

    • In vitro culture of hybrid cells derived from flounder gill cells and shrimp lymphocytes

      Abstract:The hybrid cells produced by PEG2mediated cell fusion method with gill cells from flounder , Paralichthys olivaceus , and lymphocytes from shrimp , Penaeus chinensis , were cultured in mixed medium of MEM∶MPS (1∶1) . After 5 days culture and observation , it was found that the hybrid cells cultured in the mixed medium of MEM∶MPS (1∶1) grew into monolayer and with good growing and proliferating appearance. As the concentration of MPS in the mixed medium was raised gradually, ideal hybrid cells had been screened out. By now , the obtained hybrid cells have been subcultured for 12 generations and still had very good growing and dividing appearance. Hybridoma cells acquired obvious disease appearance such as ,the cell contracted to round , disattachmentanddeath,afterinoculationwithwhitespotsyndromevirus (WSSV) ofshrimpfor 10 days, and died almost completely at the 14th day after inoculation. After 0. 3 mL culture supernatants of the cells with obvious disease appearance added into a new monolayer of normal hybrid cells , the hybrid cells acquired obvious disease appearance 5 days later , while the control cells still with normal appearance. It can be concluded that the hybrid cells obtained in this study are propably the target cells of shrimp viruses.

    • Oreochromis niloticus Comparison of genomic DNA of hybrid (♀× O. aureaus ♂) and its parents

      Abstract:Using Random Amplified Polymorphic DNA (RAPD ) technique with 20 kinds of random primers , genomic DNA of hybrid ( Oreochromis niloticus ♀×Oreochromis aureaus ♂) and its parents were analysed. The resultsshowedthatonlywiththetwokindsofprimers,OPZ -14 andOPZ -18 ,species2specific fragments were produced between parent fishes , even between different fish belonging to the same species , and some of them could be used as molecular markers to identify O. niloticus and O. aureaus. And 8 primers generate polymorphic products among hybrid fish and its parents. DNA fragments amplified from the hybrid fish were more abundant than those from its parents , which may be the result of more abundant genes in the hybrid fish , because of heredity or mutation. These results provide some molecular biological basis for species identification and heterosis effect in fish.

      Abstract:This paper reports several physiological and biochemical changes related to active oxygen injure and scavenging effect in a low temperature resistant strain Isochrysis galbana Parke 8701 under low temperature stress and investigates its low temperature tolerant mechanism in comparison with a low temperature sensitive strain I. galbana Parke 3011. The experimental results showed that : ⑴the activity of superoxide dismutase (SOD) in I. galbana Parke 8701 decreased slightly when cultured at low temperature ( 8 ℃) for a week , but still kept extremely high enzymatic activity , so it could scavenge the extra active oxygen rapidly under low temperature stress and keep a membrane lipid peroxidation in relatively lower level as well as a relatively low electrolytic leakage; ⑵the membrane lipid peroxidation level and electrolytic leakage in I. galbana Parke 3011 were significantly higher than in I. galbana Parke 8701 when stressed under low temperature for a week. This was due to the relatively low SOD activity in the former and its activity rapidly declined; ⑶in comparison with SOD , peroxidase (POD) did not play an important role in relieving the low temperature injury and raising the capacity of low temperature tolerance in these two strains of I. galbana Parke.

      Abstract:The species composition and seasonal succession of zooplankton in saline..alkaline ponds in Zhaodian Fish Farm, Gaoqing County, Shandong Province, from 5 April in 1997 to 1 September in 1998, were studied. The results indicated that salinity ranged from 1 . 36 to 20 g ..L- 1 , total alkalinity changed from 2. 4 to 7. 2 mmol..L- 1 , pH changed f rom 8 to 9, the zooplankton in saline..alkaline ponds were composed by freshwater salt.. tolerance species or halophile species, and some of them were halobiont species and which usually occurs in f reshwater. Dominant species of zooplankton were Askenasia volvox , Didinium nasutum, Lagynophrya conif era, Cyclidium citrullus, Strobilidium gyrans and Prorodon viridis ( Protozoa ) ; Brachionus plicatilis, Trichocerca pusilla, Brachionus calycif lorus , B . angularis, B . urceus, Asplanchna brightwelli , Proales simplex , Filinia cornuta and Polyarthra trigla ( Rotatoria ) ; Moina micrura, Diaphanosoma leuchtenbergiatum ( Cladocerans) ; Sinocalarus tenellus, Cyclops vicinus , Thermocyclops taihokuensis ( Copepod) . The biomass of zooplankton in the control ponds and new ponds were higher than that in other fish..culture ponds.

      Abstract:Polyculture of Chinese shrimp ( Penaeus chinensis ) with various biomass of Ulva ( Ulva pertusavar ) was conducted in ten aquaria of each 53cm @ 24cm @ 23cm from July to September in 1998. The results showed that Ulva could absorb N and P nutrients and improve water quality by decreasing the nitrogen and phosphorus loading and increase dissolved oxygen levels in the polyculture systems. The shrimp had total N and P utilization rates of 42. 7% - 212. 9% higher in the tanks with Ulva than those in the monoculture system. When the densities of shrimp and Ulva were optimal ( 320- 480 g#m- 3 and 400- 600 g#m- 3) , the oxygen produced by Ulva was suff icient for shrimp. s need without extra aeration.

    • In vivo induction of interferon-γ like activity from Ctenopharyngodon idellus by phytoagglutinin

      Abstract:An interferon factor was detected in grass carp ( Ctenopharyngodon idellus) serum, induced in vivo with phytoagglutinin by cell pathological effect inhibition technique. Study on its biochemical and biological characteristics showed that it was different f rom the interferon-A/B induced by virus. It was sensitive to heat ( 56 e ) , pH2 and 0. 1% SDS, and its activity could not be neutralized by the antibody against interferon-A/B. It proved that the inducer was mitogen but w ith non-virus character, and the production could be modulated signif icant ly by the phorbol myristate ester and interleukin-2. All these were consistent with the interferon-Cin human, which indicated it was interferon-Clike activity. The induction was influenced by the temperature, inducing time and dosage of phytoagglutinin. The activity was higher at the temperature of 25 e or 30 e and at phytoagglutinin dosage of 0. 5- 0. 8 mg per f ish than that at 8 e or 15 e and at that of 0. 1- 0. 3 mg per fish. The inducing time when the activity was the highest was 3 days at high dosage of phytoagglutinin, which was shorter than that of 5 days at low dosage. The interferon induction could be promoted by the phorbo l myristate ester and interluekin-2.

    • Polymerase chain reaction ( PCR) amplification on diagnosis of Siniperca chuatsi virus disease

      Abstract:Two RAPD fragments of Siniperca chuatsi virus ( SCV) genome DNA recovered from agrose gel were inserted into plasmid pUC19 ( called SCVE369 and SCVE450) . The sequences reported the length of cloned f ragments were 369bp and 450bp. It doesn.. t show signif icant homology sequences against to those in GenBank. According those sequence two oligonucletide primersets ( P1/ P2 and P3/ P4) were designed and used to amplify genome DNA of SCV by the method of Polymerase Chain Reaction ( PCR) . The results showed primer ( P1/ P2) could specially amplify 369bp in size.. s fragment in SCV genome. Furthermore, this primer set was used to test the DNA samples including the normal mandarin f ish, cultured f ish naturally infected w ith SCV and the fish artif icially infected by SCV. The 369bp in size PCR product was only obtained in the mandarin fish carried SCV. Primer ( P1/ P2) can been used to diagnose SCV disease of mandarin f ish.

    • Characterization of shrimp white spot syndrome virus(WSSV) infection in Cambarus proclarkii

      Abstract:Crawf ishes ( Cambarus proclarkii ) as animal were infected with white spot syndrome virus (WSSV) for studying various infection characters, including temperature, transfer route, successive infection, median lethal dose ( LD50) , immunity against virus and virus conservation etc. The results showed crawf ishes usually died in 2 - 7d after infection at the temperature of 22- 25 .. , in 2- 10d at 15- 19 .. , in 3- 13d at 8- 10 .. and 2- 6d at 30- 32 .. . Below 5 .. , the infected crawf ishes could remain active for more than one month. By injecting and oral feeding, WSSV could infected crawfishes successfully and there was no significant difference between muscle injection and hypoderm injection. Soaking with WSSV failed to infect crawfishes. Bacteria examination in healthy and diseased crawfishes showed Enterobacter cloacae in the heart and hepatopancreas multiplied 25 and 30 times respectively , indicating that bacteria successive infect ion existed during the virus infection. The LD50 was 10 - 6. 5..mL - 1 WSSV. Crawfishes which were immunized with WSSV inactivated at 56 .. for 30min could not obtain immunity protection from. WSSV f rom homogenate stored at - 30 .. for 1 year could not be recovered but f rom crawfishes stored at - 30 .. for 1 year could be recovered successfully.

    • Purification and partial characterization of serum immunoglobulin in Anguilla anguilla

      Abstract:The purification of serum immunoglobulin in European eel( Anguilla anguilla) was carried out by using ammonium sulfate precipitation followed by column chromatography. The results showed that most of the eel serum Ig was precipitated at 30% to 50% ammonium sulfated saturation. Further purification by Sephacryl- S200 showed that the Ig presented in the first protein peak , and by Sepharose 4B the eel Ig existed at the second protein peak; the purif ied product ion from Sepharo se 4B were further separated by DEAE-52 column into two f ractions, and the Ig was found in the first f raction. The antibody activity of the productions stated above was proved by ELISA and Western-blot. SDS-PAGE showed that the heavy chain and light chain of the eel serum Ig were 68kD and 26kD respect ively.

    • Effects of non-biologic environmental factors on Penaeus chinensis infected by the explosive epidemic disease virus

      Abstract:The ef fect of non-biologic environmental factors under satured dissolve oxygen concentration on Penaeus chinensis infected by explosive epidemic virus was studied. The level of salinity, temperature, pH and NH3-N was changed, according to the orthogonal table. Factors appeared different influence degree at the different stages after infection. In 3- 4 days the inf luence degree of these four factors from heavy to light was salinity, pH, temperature and NH3-N, but from the fif th day the order was changed as NH3-N, pH, temperature and salinity. After that, the level of salinity was fixed and the level of temperature, pH and NH3-N was changed , according to mixed level of orthogonal design. The result showed that the inf luence degree of three factors from heavy to light was temperature, NH3-N and pH. This result accorded with the explosive time of the shrimp disease in shrimp culture industry in China. The relationship among environmental factors, and incidence of the disease and mortality rate of Penaeus chinensis was discussed.

    • Scavenging effects of fucoidan fractions of low molecular weight extracted from Laminaria japonica on radicals of active oxygen and antioxidation in vivo

      Abstract:The scavenging effects of three fucoidan fractions ( LMWF-M, LMWF-I, and LMWF-IV) of low molecular weight extracted from Laminaria japonica on free radicals of active superoxide anio( O -.. 2 ) and hydroxyl ( ..OH) were studied by means of the chemiluminescence analysis. The experimental result s showed that three f ractions all exhibited signif icant scavenging capacity on O -.. 2 which was increased with the increase of LMWF concentration. LMWF- I containing high glucuronic acid but low sulfate content displayed the highest scavenging effect on free radicals of act ive oxygen , and its IC50 on O2 -.. was 0. 044mg..mL - 1 and 0. 062mg..mL - 1 on ..OH. Moreover, LMWF- M showed strong antioxidative activity in the experimental rat with hyperlipidemia. The content of LPO in serum and tissue was greatly reduced in hyperlipidemic rats( P < 0. 01) , while the activity of SOD was effectively enhanced ( P < 0. 01) .

    • Comparison of different degreasing technique of Pneumatophorus japonicus

      Abstract:The study focuses on the comparison between degreasing technique of chub mackerel( Pneumatophorus j aponicus) fillet and surimi w ith alkaline lipase hydrolysis and physicochemical method ( bleaching and pressure) respectively . These research result s indicated the degreasing of physicochemical method ( w ith 11. 01 of lipid residue rate based on dry weight, the below is as the same) is superior to enzyme hydrolysis ( with 19. 48% of lipid residue rate) for chub mackerel f illets, but on the contrary the enzyme hydrolysis ( with 3. 49% of lipid residue rate) is much superior to the physicochemical method ( with 7. 52% of lipid residue rate ) for chub mackerel surimi. The optimum conditions of enzyme hydrolysis are selected as follows pH from 8. 7 to 9. 2; temperature at 32 e ; alkaline enzyme solution w ith a lipase activity of 40U#mL- 1; the ratio between enzyme solut ion and surimi to be 5: 1; time of enzyme hydrolysis for 50 min. The results showed that the lipid residue rate of Chub mackerel surimi is less than 4% by using alkaline lipase and about 8% by using physicochemical method respectively under the above enzyme hydrolysis. The residual lipid rate of f illet is dif ficult to reach below 10% either by alkaline lipase or by physicochemical method.

    • Assessment of eutrophication in the north of Zhejiang coast

      Abstract:Depend on the indices of COD, T-N, PO4-P and chlorophy11-a, the eutrophication in the north of Zhejiang coast was assessed by the method that associated with weighing distance and subordinative degrees of fuzzy set theory. In the course of the assessment, the significance-weighing vector was determined by the modif ied hierarchy analyzing method and the signif icance-compared matrix of the assessment indices was adjusted. The assessment results were given in two ways of subordinative and accumulative methods. The results showed that the area with middle and middle-high eutrophication were 76. 6% of the researched area, while the area with lower and higher than that being 16. 6% and 6. 7% respectively. The different arranges of assessment indices influenced the f inal assessment results, and the accumulative method was of higher exactitude than the subordinative method for describing status of eutrophication. The further research needs to be carried out on the relativity between ertrophication and algae bloom.

      Abstract:11 968 Sparus marcrocephalus with average body length of 60 .2mm were tagged and released in Daya Bay during 1997 .955 Sparus marcrocephalus were recaptured and the recapture rate was 8 .0 %.The most part of them distributed in the shallow waters along the coast of the top Bay , only few appeared in the ialands of the middle Bay .The tagged fish were well growing , the average body length of them had got to 235mm after 240 days .
